Haptoglobin Electrochemical Diagnostic Method for Subclinical Mastitis Detection in Bovine Milk

چکیده

This work proposes an outstanding screening magneto-bioassay for the early identification of bovine subclinical mastitis. Haptoglobin (Hp) was used as a promising biomarker diagnosis To this end, novel chitosan-modified magnetic particles, coated with haemoglobin-modified polyaniline (MNPs@Chi/PANI-Hb), have been integrated in sensitive electrochemical Hp bioassay. Haemoglobin bioreceptor due to its high affinity against Hp. The appropriateness their synthesis and modifications were demonstrated by XRD, FT-IR spectroscopy, thermogravimetry N2 adsorption–desorption analyses. After optimization experimental parameters main analytical features obtained showing excellent performance. biosensor milk matrix presented dynamic range 0.001 0.32 μg mL−1 detection limit 0.031 mL−1, value much lower than normal values milk, making it highly suitable such determinations. Finally, real samples, from local dairy farmers, analysed bioassay compared commercial ELISA kit detection. Milk samples correctly classified “acceptable” or “unacceptable” considering concentration proposed bioassay, confirming predictive capacity mastitis diagnosis.

Characterization of Haptoglobin Isotype in Milk of Mastitis-Affected Cows

Haptoglobin is a major acute phase protein in bovines and reportedly increases in serum and milk whey during mastitis, highlighting its potential as a diagnostic biomarker. Since haptoglobin is known to undergo tissue specific glycosylation resulting in different isoforms, this study was undertaken to characterize the isoforms of haptoglobin. Milk whey fraction and serum obtained from animals w...

Ultrasonography of the supramammary lymph nodes for diagnosis of bovine chronic subclinical mastitis

Currently, somatic cell count (SCC) and bacterial culture are considered as the gold standard of detecting subclinical Mastitis. Mastitis leads to proliferation of lymphocytes in the supramammary lymph nodes and subsequent enlargement of ipsilateral lymph node. Ultrasonography can be used to survey these changes. A portable ultrasound machine with a 2-5 MHz convex transducer was used to identif...

Biosensor assay for determination of haptoglobin in bovine milk.

Despite more than 30 years of research into mastitis diagnostics, there are few alternatives to the somatic cell count (SCC) in practical use for identification of cows with subclinical mastitis. Mastitis is not only an animal welfare problem, but also affects the yield, composition and technological properties of milk. Hence, dairy cooperatives give farmers a premium quality payment to encoura...

Assessment of Lactate Dehydrogenase Enzyme Activity in Milk as a Marker for Detection of Subclinical Mastitis

Sub clinical mastitis is responsible for heavy economic losses throughout the world in dairy sector. The objective of present study was to investigate lactate dehydrogenase (LDH) enzyme activity in milk for detection of subclinical mastitis in buffaloes Milk samples from 60 buffaloes, including 40 subclinical infected and 20 healthy were subjected to the estimation of LDH using a LDH activity a...

Screening method for Staphylococcus aureus identification in subclinical bovine mastitis from dairy farms

BACKGROUND Staphylococcus aureus is one of the most important contagious bacteria causing subclinical bovine mastitis. This bacterial infection is commonly identified by determine the pathogen in bovine milk samples through conventional technique including coagulase test. However, this test has several disadvantages as low sensitivity, risk of biohazard, cost expensive, and limited preparation ...
